Single-Cell RNA Sequencing Maps Immune Cell Heterogeneity in Mice with Allogeneic Cardiac Transplantation

چکیده
Objective: Immune cells play important roles in mediating allograft rejection and tolerance after cardiac transplantation. However, immune cell heterogeneity at the single-cell level, how states shape transplantation immunity, remain incompletely characterized. Methods: We performed RNA sequencing (scRNA-seq) on LNs from a mouse syngeneic allogeneic model. Nine T clusters were identified through unsupervised analysis. Pathway enrichment analysis was used to explore functional differences among subpopulations characterize metabolic of cells. Results: comprehensively determined transcriptional landscape cells, particularly their transcriptomes during On basis molecular properties, we also types associated with transplantation-associated processes, including cytotoxic CD8 + activated conventional CD4 dysfunctional Tregs. further elucidated contribution JunB induction Th17 differentiation restriction Treg development, that HIF-1a participates metabolism function. Conclusions: present first systematic variation within population, providing new insights for development novel therapeutic targets rejection.

A Graph-Based Clustering Approach to Identify Cell Populations in Single-Cell RNA Sequencing Data
Introduction: The emergence of single-cell RNA-sequencing (scRNA-seq) technology has provided new information about the structure of cells, and provided data with very high resolution of the expression of different genes for each cell at a single time. One of the main uses of scRNA-seq is data clustering based on expressed genes, which sometimes leads to the detection of rare cell populations. ...

Linezolid (LZD), severed as the first oxazolidinone antibiotic, was active against multidrug-resistant gram-positive strains. LZD can induce thrombocytopenia, anemia and leukocytopenia. Currently, reports on pure red cell aplasia (PRCA) cases induced by LZD are relatively rare [4-7]. In this paper, we reported a patient with PRCA twice induced by LZD. A 37-year-old man was diagnosed with myelody...

متن کاملI-13: Transcriptome Dynamics of Human and Mouse Preimplantation Embryos Revealed by Single Cell RNA-Sequencing
Background: Mammalian preimplantation development is a complex process involving dramatic changes in the transcriptional architecture. However, it is still unclear about the crucial transcriptional network and key hub genes that regulate the proceeding of preimplantation embryos. Materials and Methods: Through single-cell RNAsequencing (RNA-seq) of both human and mouse preimplantation embryos, ...
